Abstrakty
Smak gorzki wielu produktów jest składową kilku gorzkich związków chemicznych, a nasilenie tej cechy smakowej wyraźnie zależy od stopiona przetworzenia produktu, pochodzenia i jego jakości. Substancje goryczkowe na trwałe zakorzeniły się w składzie produktów spożywanych przez ludzi. Cechy funkcjonalne wraz z tzw. "atrakcyjnością sensoryczną" decydują w największym stopniu o popycie na dany produkt. Bardzo istotna dla badań nad smakiem gorzkim jest też problematyka preferencji i akceptacji tego smaku w produktach spożywczych. Prace badawcze naszego zespołu przyczyniły się również do wymodelowania matrycy molekularnej receptora gorzkiego, która jest specyficzną supermolekułą niezbędną dla określenia interakcji receptor - związek gorzki. W prezentowanych badaniach nad smakiem gorzkim pochodnych chromonów i metyloksantyn, metoda dokowania w matrycy molekularnej oraz metoda QSAR stanowi integralną całość. Zastosowanie metody dokowania w matrycę molekularną wraz z metodą QSAR pozwoliło na wyznaczenie dobrej liniowej korelacji pomiędzy smakiem gorzkim i strukturą badanych alkaloidów metyloksantynowych i chromonów. Wyprowadzenie takiego równania smakowego dla chromonów i alkaloidów metyloksantynowych może znacznie uprościć poszukiwania innych związków zaliczanych do tego szeregu i posiadających smak gorzki.(oryg. streszcz.)
Bitter taste of many products follows from the presence of a few bitter compounds and the bitter taste intensity significantly depends on the degree of the product processing, its origin and quality. Bitter taste substances have found permanent application in the composition of food products. The functional characteristics together with the so-called sensory attractiveness determine to the greatest degree the demand for a given food product. The problems very important in the study of bitter taste perception are man's preferences and acceptance of bitter taste in food products. The research work of our group has contributed to development of a model molecular mould of the bitter taste receptor, being a specific supermolecule necessary for determination of the interaction between the bitter taste carrying compound and the receptor. In the study on the bitter taste of chromones derivatives and methylxanthine alkaloids the methods of docking into the molecular mould and QSAR have been used as complementary. The method of docking into the molecular mould in combination with the QSAR calculations has permitted determining a good linear correlation between the bitter taste and the structure of the chromones and methylxanthine alkaloids studied. The equation derived for methylxanthine and chromones derivatives can simplify the search for other bitter taste compounds. (original abstract)
