W artykule przedstawiono wybrane, powszechnie stosowane metody oznaczania aktywności przeciwutleniającej oraz wyniki badań, przeprowadzonych w Katedrze Instrumentalnych Metod Oceny Jakości, nad oceną czynników decydujących o aktywności przeciwutleniającej TEAC ważnych grup związków polifenolowych występujących powszechnie w żywności pochodzenia roślinnego oraz nad mechanizmem ich przeciw-utleniającego działania. Wyniki badań eksperymentalnych porównano z obliczonymi teoretycznie parametrami molekularnymi związków polifenolowych. Na podstawie wyznaczonych zależności pomiędzy strukturą i aktywnością fenolokwasów, hydroksyflawonów, antocyjanów i katechin wykazano, że ważnymi czynnikami decydującymi o aktywności przeciwutleniającej związków polifenolowych jest nie tylko liczba i rozmieszczenie grup hydroksylowych, lecz również stan ich protonacji, zależny od pKa grup hydroksylowych, pH środowiska i efekt addytywny niezależnych, aktywnych przeciwrodnikowo ugrupowań występujących w cząsteczkach polifenoli. The study presents commonly used methods for determination of the antioxidant activity and evaluates various factors influencing the TEAC antioxidant activity of these important classes of phenolic antioxidants, commonly present in food of plant origin, giving more insight into the mechanism of their antioxidant activity. Experimental data were compared to theoretically calculated molecular parameters. Based on the structure - activity relationships obtained for phenolic acids, hydroxyflavones, anhocyanins and cat-echins, it was shown that important factors determining the antioxidant activity of phenolic compounds are not only the number and position of the hydroxyl moieties but also their protonation state, influenced by the pKa of the hydroxyl moieties, the pH of the medium, influenced by the pKa of the hydroxyl moieties, and the additive effect of independent radical scavenging moieties present in their molecules.
