Salmonella sp. jest jedną z głównych przyczyn zatruć pokarmowych. Z uwagi na czasochłonność standardowej metody wykrywania tego patogenu w żywności istnieje potrzeba doskonalenia metod alter-natywnych. W badaniach zastosowano fluorescencyjną hybrydyzację in situ (FISH) z użyciem sondy Sal3 do wykrywania obecności Salmonella sp. w mięsie drobiowym (zaszczepionym Salmonella sp. oraz innymi pałeczkami z rodziny Enterobacteriaceae) i porównano ją z immunodiagnostyczną metodą enzymo- immunofluorescencyjną (VIDAS® SLMX) oraz metodą wg PN-ISO-6579. Metodami: FISH i VIDAS nie otrzymano wyników fałszywie negatywnych. Metodą VIDAS uzyskano jednak kilka wyników fałszywie pozytywnych. W metodzie FISH nie zaobserwowano hybrydyzacji ze szczepami innymi niż Salmonella sp. Wyniki wskazują na możliwość zastosowania FISH jako szybkiej metody wykrywania Salmonella sp. w mięsie drobiowym. Salmonella sp. is one of the main causes of food poisoning. Since the standard method for detecting that pathogen in food is time-consuming, it is necessary to perfect alternative methods. In the research study, a fluorescent in situ hybridization method (FISH) was utilized with the use of a Sal3 probe to detect Salmonella sp. in chicken meat (inoculated with Salmonella sp. and with other Enterobacteriaceae rods) and compared with an Enzyme-Linked Fluorescent Immunoassay-Based Method (VIDAS® SLMX) and with an ISO method. While using the FISH and VIDAS methods, no false negative results were obtained. With the use of the VIDAS method, several false positive results were obtained. In the FISH method, no hybridization was reported to any strains other than Salmonella sp. The results obtained indicate that the FISH method could, possibly, be utilized as a rapid screening method for detecting Salmonella sp. in chicken meat. (original abstract)
