Influence of Alcohols and Detergents on Divercin Aggregation

Stosując metodę filtracji membranowej stwierdzono, że diwercyna wykazuje tendencję do silnej agregacji w kompleksy zawierające ponad 25 monomerów. Agregaty diwercyny ulegały częściowemu rozszczepieniu pod wpływem etanolu, izopropanolu, SDS, Nonidetu P-40 oraz Tweenu 80. Najsilniejsze właściwości dezagregujące posiadał SDS. Spośród badanych alkoholi i detergentów, jedynie Tween 80 mógłbyć dodawany bezpośrednio do pożywki, gdyż nie wykazywał toksycznego działania na kulturę Carnobacterium divergens AS7. Obecność tego związku w pożywce hodowlanej zwiększała aktywność diwercyny, skutecznie ograniczała jej agregację i ułatwiała ultrafiltrację. Biorąc pod uwagę możliwość zastosowania tej bakteriocyny w przemyśle spożywczym, do dezagregacji diwercyny zalecany jest alkohol etylowy. (abstrakt oryginalny)
Based on the divercin transfer through ultrafiltration membrane, it was showed that this bacteriocin tents to form large aggregates with the molecular weight over 100 kDa, contains over 25 monomers. The aggregates were partially depolymerised by ethanol, isopropanol, SDS, Nonidet P-40 and Tween 80. The greatest depolymerization of divercin aggregates was observed when SDS was used. Among alcohols and detergents tested, only Tween 80 can be introduced directly into Carnobacterium divergens AS7 culture because of its non toxic effect on the bacteria cells. The presence of Tween 80 increased divercin activity, limited divercin aggregation and make easy its ultrafiltration. Taking into account the application of divercin in food industry, ethanol can be suggested for disaggregating of bacteriocin macromolecules. (original abstract)
