Computer-Based Prediction of the Bitter Taste of N-Alkyl and N-Acyl Cytisine Derivatives

The molecular mould of bitter taste is a kind of universal "molecular form" of the bitter taste receptor as it imposes a certain specific spatial orientation of the agonist in the receptor. The computer projections of cytisine derivatives into the molecular mould permitted to rank having accord to increasing values of the bitter taste indices this compounds. This observation confirms, that with increasing size of the substituent at N12 the taste indices characterising these compounds should decrease. The results obtained have confirmed, that at the border of the sectors A and B of the molecular mould are localised bipolar subsite of electron-donor and proton-donor character. (original abstract)
Prognozowanie smaku gorzkiego związków organicznych wymaga rozpoznania sposobu wiązania się struktur gorzkich ligandów w jamie receptorowej. Identyfikacje tych oddziaływań przeprowadziliśmy, używając dokowania struktur gorzkich ligandów smakowych do wnętrza matrycy molekularnej. Matryca molekularna to uniwersalna "forma molekularna" receptora smaku gorzkiego, wyznaczona za pomocą wzajemnego nałożenia struktur gorzkich ligandów. Cytyzyna naturalnie występująca w nasionach Laburnum cmagyroides wraz z syntetycznymi analogami zaliczana jest do grupy alkaloidów chinolizydynowych. Istotą różnic smakowych badanych alkaloidów jest budowa i dostępność grupy aminowej lub amidowej (atomy azotu Ni i Nn). Projekcje komputerowe pochodnych cytyzyny w matrycę molekularną wykazały, że cytyzyna jako jedyny z badanych związków oddziałuje z obszarami elektrofilowymi jak i nukleofilowymi matrycy. Tym samym cytyzyna stymuluje kilka obszarów rozpoznania smakowego i jest najsilniejszym gorzkim ligandem w badanej serii związków. Zupełnie odmienna sytuacja ma miejsce w przypadku benzylo propionylocytyzyny, która oddziałuje tylko ugrupowaniem amidowym co sprawia, że jest praktycznie nie gorzka. Pochodne metylowa, acetylowa i propionylowa cytyzyny stymulują po dwa nukleofilowe obszary aktywne i tym samym ich indeksy smaku gorzkiego mają wartości pośrednie. Prognozowany smak badanych alkaloidów potwierdzają wyniki przeprowadzonych analiz sensorycznych. Uzyskane wyniki dowodzą, że na pograniczu sektorów A i B matrycy molekularnej jest zlokalizowany dwubiegunowy subregion o charakterze nuklo(elektofilowym). Prezentowane wyniki badań umożliwią znaczne skrócenie analiz smakowych związków gorzkich będących składnikami wielu produktów spożywczych, leków i suplementów diety. (abstrakt oryginalny)
